MDB-38. COMPUTATIONAL DRUG SENSITIVITY PREDICTS MEDULLOBLASTOMA SUBGROUP-SPECIFIC THERAPEUTICS

چکیده

Abstract Medulloblastoma is the most common malignant pediatric brain tumor. Tumors are typically characterized as Group 3, 4, SHH, or WNT. Current standard-of-care includes surgery, radiation, and chemotherapy; however, treatment response prognosis vary widely between subgroups. Additionally, surviving children frequently suffer from lifelong neurocognitive deficits. Therefore, novel therapeutic options urgently needed. Despite extensive characterization of medulloblastoma tumors into four molecular subgroups, few subgroup specific therapies have advanced to clinic. To address this, we developed a platform called DrugSeq, which predicts drug sensitivities in patients can stratify by subgroup. DrugSeq also identifies key pharmacotranscriptomic differences primary recurrent tumors. We first calculated disease signatures for each patient normalizing gene expression low grade glioma samples posterior fossa. Using previously transcriptional consensus (TCSs) that represent affect has on across panel cancer cell lines, then discordance signature TCS. Finally, using an ANOVA analysis identified drugs predicted differentially target Among our top anti-cancer compounds found several kinase inhibitors, bromodomain psychiatric with known penetrance. show distinct sensitivity predictions newly diagnosed tumors, such BET inhibition 3 4 despite no Future studies larger datasets may be able further subdivide age features within Collectively, identify facilitate stratification clinical trials, leading more successful targeted therapies.

Subgroup-specific prognostic implications of TP53 mutation in medulloblastoma.

PURPOSE Reports detailing the prognostic impact of TP53 mutations in medulloblastoma offer conflicting conclusions. We resolve this issue through the inclusion of molecular subgroup profiles. PATIENTS AND METHODS We determined subgroup affiliation, TP53 mutation status, and clinical outcome in a discovery cohort of 397 medulloblastomas. We subsequently validated our results on an independent ...

Striatal dopamine predicts outcome-specific reversal learning and its sensitivity to dopaminergic drug administration.

Individual variability in reward-based learning has been ascribed to quantitative variation in baseline levels of striatal dopamine. However, direct evidence for this pervasive hypothesis has hitherto been unavailable. We demonstrate that individual differences in reward-based reversal learning reflect variation in baseline striatal dopamine synthesis capacity, as measured with neurochemical po...

Medulloblastoma subgroup-specific outcomes in irradiated children: who are the true high-risk patients?

BACKGROUND The advent of integrated genomics has fundamentally changed our understanding of medulloblastoma. Although survival differences exist among the 4 principal subgroups, this has yet to be elucidated in a North American cohort of irradiated patients. METHODS Ninety-two consecutive patients between the ages of 3 and 17 treated with surgery, craniospinal irradiation, and chemotherapy we...

MicroRNA–mRNA expression profiles associated with medulloblastoma subgroup 4

Purpose Medulloblastoma (MB), the most common malignant brain tumor in children, is divided into four tumor subgroups: wingless-type (WNT), sonic hedgehog (SHH), Group 3, and Group 4. Ideally, clinical practice and treatment design should be subgroup specific. While WNT and SHH subgroups have well-defined biomarkers, distinguishing Group 3 from Group 4 is not straightforward. MicroRNAs (miRNAs)...

MR Imaging Characteristics of Wingless-Type-Subgroup Pediatric Medulloblastoma.

BACKGROUND AND PURPOSE "Transcriptionally different" medulloblastoma groups are associated with specific signaling pathway abnormalities; hence, they may present with distinct imaging manifestations. In this study, we sought to describe the MR imaging features of wingless-type-subgroup medulloblastomas with embryologic correlations. MATERIALS AND METHODS Pre- and postoperative imaging studies...
